After graduating high school, he went on to play at the University of Louisville, where he helped lead the team to a National Championship in 1986. His professional playing career spanned over a decade, during which he played in the NBA and internationally.

After retiring from professional basketball, Payne transitioned to coaching. He spent several years as an assistant coach at the University of Oregon and the New York Knicks before returning to his alma mater, Louisville, as an assistant coach in 2010. In 2022, he was promoted to head coach, making him the first African American to ever lead the Louisville Cardinals program.
