Majodina was born in Port Elizabeth, South Africa, on April 21, 1957. She began her career as a singer in the 1970s, and released her debut album, The Magic of Pemmy Majodina, in 1981.
In the 1980s, Majodina began acting in theater and film. She made her television debut in the 1988 series Egoli: Place of Gold.
In 1994, Majodina joined the cast of Generations. She played the role of Ma Agnes for over 20 years, becoming one of the most beloved characters on South African television.
